The topic: Exploiting High Resolution Accurate Mass QTof MSMS for the Identification of Unknown Analytes in Biomedical Device Leachates and Extracts will cover ISO 10993 leachables and extractables testing.

Abstract: Small molecule structural elucidation aided by high resolution mass spectrometry is built on a foundation of high accuracy mass measurements. Accurate mass data allow elemental composition searching and isotopic pattern matching of molecular ion masses which can often lead to the identification of a specific elemental composition for the unknown molecule consistent with the data.

Elemental composition is only the first step in structural elucidation, as one elemental composition represents a significant number of viable chemical structures. Consequently, the next significant challenge is to trim the viable structure list and this requires covalent bond structure characterization.  This is aided by MSE, an optimized MSMS acquisition mode that is unique to the Waters Corporation Xevo G2-S QTof mass spectrometer.  MSE yields complete fragment ion information for every significant chromatographic peak in one analytical run.  Experimental fragment ion data require expert interpretation.

The utility of Mass Frontier, a Thermo Scientific fragment analysis software, will be discussed. Mass Frontier is an in silico-based, i.e.: predictive, fragmentation elucidation software that generates structures of expected fragments from a given precursor ion. Successful application of the work flow discussed above will be demonstrated through three biomedical device case studies involving chemical identification of unknown analytes detected in biomedical device leachates, a challenge typically encountered in ISO 10993 testing.

Marie Dvorak Christ, Ph.D., Chemistry, University of California, Irvine, the founder of Mass Spec Lab, is a leader in the development of accurate mass TOF LC/MS and GC/MS methods for extractables and leachables analysis applied to biomedical devices, pharmaceuticals and other allied products.

With fifteen years of experience at Invision Biomedical, Dr. Christ has earned an exceptional reputation for successfully solving difficult chemical problems encountered in biomedical device product development. Dr. Christ holds several patents in the area of ophthalmic materials and has had extensive experience in project management, materials development, and L&E materials/device testing.

Prior to founding Invision Biomedical and Mass Spec Lab, Dr. Christ worked in Professor David Brant’s group at UCI where her doctoral work focused on the NMR and MS structural elucidation of biologically active sulfated polysaccharides fundamental to species specific fertilization in sea urchins. Dr. Christ’s postdoctoral academic research at the UCSD Cancer Center with Dr. Ajit Varki and at UCR with Professor Dallas Rabenstein focused on the structural characterization and chemical analysis of peptides, glycans and glycoconjugates utilizing both mass spectrometry and NMR.
